History:

Palmitoyl tripeptide-5 (Palmitoyl-Lysyl-Valyl-Lysine, Pal-Lys-Val-Lys-OH) is also called "tripeptide" or "collagen peptideCOLL-O2 ", the most widely used cosmetic peptide. Palmitoyl tripeptide-5 belongs to the signal peptide class and can promote the synthesis of collagen (I type, II types, and IV type) and the production of extracellular matrix protein by increasing the activity of stromal cells to make the skin looks young and elastic. Collagen can be added to cosmetics to help skin supplement collagen, improve facial elasticity and remarkably reduce facial wrinkles. Meanwhile, the skin care product has the characteristics of enhancing cell activity, increasing the skin’s water content, enhancing the skin luster, and improving the skin color.


In the prior art, more research reports about applying palmitoyl tripeptide-5 exist. As a result of research on the whitening efficacy and action mechanism of palmitoyl tripeptide-5 in the literature [ daily chemical industry, 2018,48(3),166-171], the palmitoyl tripeptide-5 can inhibit the generation of melanin in melanocytes by inhibiting tyrosinase activity and reducing mRNA levels of microphthalmia-associated transcription factor (MITF) and tyrosinase and can be applied to the field of cosmetics as a whitening additive. Chinese patent publication No. CN108619047A discloses an anti-aging cosmetic formulation and a preparation method, wherein one of the anti-aging components is palmitoyl tripeptide 5. Chinese patent with publication No. CN 107669564A discloses a skin care composition with repairing and regenerating, moisturizing, and delaying senility, wherein palmitoyl tripeptide-5 is one of the main components. Many reports of related research need to be listed here.

However, the literature on preparing palmitoyl tripeptide-5 in the prior art is scarce, and the practice of palmitoyl tripeptide-5 by a solid phase synthesis method is mentioned in U.S. Pat. No. 5, 2007099842, 1, but various protecting groups and deprotecting reagents are required, and the final yield is only 19%. Besides the method, this is the only method for preparing the palmitoyl tripeptide-5 reported at present. (CN201910789414.XA, 2020)
